Biosynthesis of Pyoluteorin: A Mixed Polyketide-Tricarboxylic Acid Cycle Origin Demonstrated by [l,2 -13C2]Acetate Incorporation

چکیده
Pyoluteorin (1) is an antibiotic and phytotoxic metabolite of several Pseudomonas spp. [1 — 3]. Its biosynthesis had not been studied previously but consideration of its structure suggested that it is a tetraketide with proline (or equivalent) as the starter unit. This has now been confirmed by an incorpora tion experiment employing [l,2 -13C2]acetate as pre cursor and analysis of the isolated labelled product by 13C nuclear magnetic resonance spectroscopy (13C NMR). The basis for the experiment was the expectation that acetate would be incorporated into the resorcinol moiety by the usual acylphloroglucinol route (Scheme 1) while the acyl moiety, i.e ., the dichloropyrrole carboxylic acid residue, would be derived from the tricarboxylic acid (TCA) cycle as in Scheme 2. With the assumption that only the first two turns of the cycle would make a significant contribution, in tact acetate units would be incorporated into two bonds in this fragments, C -l, -2 and C-4, -5, which would be revealed by the presence of the characteris tic satellite patterns flanking the normal singlets for these four carbons in the 13C NMR spectrum. Biosynthetic studies on some polyene mycotoxins
Citreoviridin, asteltoxin and aurovertin B are members of a group of mycotoxins which act as inhibitors of oxidative phosphorylation in rnitochondria. The biosynthetic origin of the carbon skeleton of these metabolites was studied by incorporation of 13C—labelled precursors: [l—'3C]—, [2—'3C]—, [l,2—'3C2]—, [l—'3C,2—2H3—, and 12—'3C,2—2H3]acetate, (25)—[methyl— 13C ]methionine and [1— 3C]— and ...
متن کاملThe Incorporation of [1,2-13C2] Acetate into Pisatin to Establish the Biosynthesis of Its Polyketide Moiety *
Biosynthesis, 13C-NMR, Phytoalexins, Pisatin The biosynthesis of the polyketide moiety in pisatin has been examined by incorporation ex periments with [1,2-13C2] acetate and 13C-NMR measurements. The results establish that the C-5 carbonyl group in the enzyme-bound, open-chain polyketide precursor is reduced before cyclization to form the aryl ring. The specific incorporation attained in these...
متن کاملCharacterization of the pyoluteorin biosynthetic gene cluster of Pseudomonas fluorescens Pf-5.
Ten genes (plt) required for the biosynthesis of pyoluteorin, an antifungal compound composed of a bichlorinated pyrrole linked to a resorcinol moiety, were identified within a 24-kb genomic region of Pseudomonas fluorescens Pf-5. The deduced amino acid sequences of eight plt genes were similar to the amino acid sequences of genes with known biosynthetic functions, including type I polyketide s...
متن کاملAmino acid biosynthesis in mixed rumen cultures.
Mixed rumen micro-organisms, maintained in continuous culture readily incorporated labelled HCO3- and acetate into amino acids. Labelled propionate, in contrast, was utilized only for isoleucine biosynthesis, but failed to label other amino acids to any significant extent. Evidence was obtained showing that in these mixed, i.e. symbiotic, cultures foward tricarboxylic acid-cycle reactions only ...
متن کاملIncorporation of acetate carbon into glucose by liver slices from normal and alloxan-diabetic rats.
In a previous report (1) we demonstrated the incorporation of palmitic acid carbon into glucose in the intact rat and estimated the quantitative significance of this process in the normal and alloxan-diabetic animal. Even though the values obtained represented the lower limit of the extent of this process, the results indicated that conversion of fatty acids to carbohydrate is a demonstrable me...
